This work was carried out to demonstrate the motility change in isolated perfilsed small intestinal loops of rats treated by melatonin. Assessment of frequency of contractions ( wavelmin) , peak tension (PT-gm), time to peak tension (TPT-sec) and half relaxation time (1/2 RT- sec~ under hasal condition and after melatonin administration. Male albino rats were used and classified into in vivo melatonin-treated rats for 2 weeks in (a) small dose ( O.05mglkg bw and (b) in a high dose ( 1mglkg bw. ). Also, studv of melatonin in vitro was carried Out in 2 doses a small dose(50ug) and a high dose(500ug) . The results obtained illustrated that melatonin in small dose significantly increased the frequency and peak tension of intestinal conlractions both in vivo and in vitro .. Also .addition of melatonin in a small dose in vitro immediatelv afier atropine significantly enhanced the intestinal contractions as regards to frequency of slow and fast waves and peak tension. On the other hand, melatonin in a high dose either in vivo or in vitro significantlv diminished the Fequency of intestinal contractions and peak tension. However, the changes in duration of TPT and 1/2 RT in melatonin-treated groups in a small dose and a high dose both in vivo and in vitro were statisticallv insignificant . It could be concluded that melatonin modulate the intestinal motility as it enhanced the intestinal tone and rhythm in low therapeutic level while it tends to attenuate them in the high dose.
